Simple bi-directional MSA setup (not replicating DDL but
manually applied at the replicate) where the replicate is at
the DR site. Though replication is also setup from the DR
site to the primary the replication agents are not started
until it starts to function as the primary after a switch.

Primary = PDS.dbname
Replicate = RDS.dbname

Should dsi_replication be set to off for both connections?

do you mean your replicate database repagent is always down and you will
enable it only when you switch your apps to point to replicate ?
If that is the case this parameter should be switched off for both the
connection. This is what manual says for this parameter...
"When dsi_replication is set to "off," the DSI executes set replication off
in the database, preventing ASE from adding replication information to log
records for transactions that the DSI executes."
